A recently granted patent (Publication Number: US11655235B2) discloses a compound of Formula 1, along with its stereoisomers, mixtures of stereoisomers, and pharmaceutically acceptable salts. The compound has various structures, including Formula 1a, 1b, 1c, 1d, and 1e. In Formula 1a, R5 can be either hydrogen or a C1-3alkyl group. The compound can also have R3 groups selected from a group consisting of —NR4R5, —OR4, —C(O)NHR4, —NHC(O)(CH2)mR4, and —NHS(O)2R4, where R4 is a phenyl substituted with 1 to 3 Z groups. The Z groups can be hydroxy, amino, halogen, cyano, nitro, carboxylic acid, C1-5alkyl optionally substituted with 1 to 3 halogens or amino, C2-6alkenyl, C2-6alkynyl, C1-5alkoxy, C1-5alkylthio, mono- or di-C1-5alkylamino, piperazinyl optionally substituted with acetyl, C1-5alkylsulfonylamino, C1-5alkylcarbonylamino, aminosulfonyl, aminocarbonyl, C1-5alkylaminocarbonyl, phenyl, phenoxy, benzyl, benzoyl, phenylaminocarbonyl, pyrazolyl, and benzoxazolyl. Additionally, R4 can be a heterocycle selected from benzo[d][1,3]dioxolyl, dihydrobenzo[1,4]dioxinyl, 2,3-dihydrobenzofuranyl, 2-oxo-1,2-dihydroquinolinyl, and 2-oxo-1,2,3,4-tetrahydroquinolinyl, optionally substituted with 1 to 3 Z groups. Other R4 groups include naphthyl optionally substituted with 1 halogen, and heteroaryl groups such as pyridinyl, pyrimidinyl, pyrazinyl, imidazolyl, pyrazolyl, triazolyl, oxazolyl, thiazolyl, furanyl, pyrrolyl, thiophenyl, quinolinyl, isoquinolinyl, chromenonyl, quinazolinyl, benzoxazolyl, benzofuranyl, benzothiophenyl, indolyl, indolinyl, benzimidazolyl, benzoxazolyl, benzothiazolyl, indazolyl, furo[3,2-c]pyridinyl, thieno[2,3-d]pyrimidinyl, thieno[3,2-c]pyridinyl, isoxazolyl, acridinyl, and naphthyridinyl, optionally substituted with 1 to 3 Z groups. The patent also covers compounds with R3 as a 5- to 12-membered heteroaryl optionally substituted by 1 to 3 Z groups, or as a 3- to 12-membered non-aromatic heterocycle containing nitrogen, optionally substituted by 1 to 3 Z groups. The compound can be used in pharmaceutical compositions with pharmaceutically acceptable excipients. It can also be used to inhibit fibroblast activation protein, treat nonalcoholic hepatosteatosis (NASH), diseases mediated by fibroblast activation protein, cancer, and to inhibit degradation of fibroblast growth factor-21. The diseases mediated by fibroblast activation protein include lipid disorders, diabetes, liver conditions, gastrointestinal conditions, inflammatory disorders, obesity, infections, hyperproliferative disorders, and cardiovascular conditions.
