A recently filed patent (Publication Number: US20230201420A1) describes a three-dimensional, engineered, biological skin tissue that can be cultured for at least 12 days. The skin tissue consists of a dermal layer and an epidermal layer. The dermal layer contains dermal fibroblasts, while the epidermal layer contains keratinocytes. Within 12 days of culture, the skin tissue develops multiple layers of differentiated keratinocytes.

The patent claims that the dermal fibroblasts in the dermal layer are primarily human fibroblasts. Additionally, the keratinocytes in the epidermal layer are primarily human keratinocytes. The epidermal layer may also include melanocytes, and the ratio of keratinocytes to melanocytes is between 90:10 and 99:1. The skin tissue may also contain organoids, such as sebocytes, glandular cells, or follicle cells, in the dermal layer and/or the epidermal layer. Furthermore, the skin tissue may include cancer cells.

Importantly, the skin tissue described in the patent does not have mature tissue innervation, perfusable lymphatic tissue, or perfusable vasculature. It is also free of pre-formed scaffold, indicating that it is a self-supporting structure. The epidermal layer is in continuous contact with the dermal layer, and its thickness ranges from about 20 µm to about 500 µm, with a preferred thickness of about 150 µm. The dermal layer, on the other hand, is about 10 µm to about 1000 µm thick, with a preferred thickness of about 500 µm.

The patent also mentions an array comprising multiple skin tissues, which can be used for in vitro assays, drug-screening assays, or cosmetic assays. Each skin tissue in the array is the same as the one described in claim 131.

In addition to the above-described skin tissue, the patent also includes two alternative embodiments. One embodiment includes a hypodermal layer, consisting of endothelial cells, in contact with the dermal layer and the epidermal layer. The other embodiment includes a basal layer, consisting of basal keratinocytes, in contact with the dermal layer and the epidermal layer. Both of these embodiments also result in the development of multiple layers of differentiated keratinocytes within 12 days of culture.
